Recombinant Canine Interleukin-8/CXCL8 (Canine IL-8/CXCL8)_C230270
Interleukin-8 (IL-8), also known as CXCL8, GCP-1, and NAP-1, is a widely expressed proinflammatory member of the CXC family of chemokines. Near its N-terminus, this 8-9 kDa chemokine contains an ELR motif which is important for its angiogenic properties. CXCL8 can associate into a homodimer or a heterodimer with CXCL4/PF4, and it can also interact with matrix and cell surface glycosaminoglycans. Mature canine CXCL8 shares 87%, 69%, and 82% amino acid (aa) sequence identiity with feline, human, and porcine CXCL8. There is no CXCL8 gene counterpart in rodent. N-terminal truncation of CXCL8 by multiple proteases generates a range of shorter forms. The bioactivity of CXCL8 is regulated by these truncations, by CXCL8 citrullination at Arg5 (N-terminal to the ELR motif), and by the decoy receptor DARC. CXCL8 effects are mediated through CXCR1/IL-8 RA, which is also used by CXCL6, and through CXCR2/IL-8 RB, which is used by multiple CXC chemokines. These receptors associate into functional homodimers and heterodimers with each other. Through both CXCR1 and CXCR2, CXCL8 promotes neutrophil adhesion to the vascular endothelium and migration to sites of inflammation. It triggers the antimicrobial activation of neutrophils through CXCR1. CXCL8 also binds to Serpin A1/alpha-1 Antitrypsin, and this prevents CXCL8 interaction with CXCR1. CXCL8 is upregulated in atherosclerotic lesions and other cardiac pathologies where it exacerbates inflammatory tissue damage. In addition, it induces VEGF expression, vascular endothelial cell proliferation, angiogenesis, and tumor cell invasiveness. Recombinant Canine Tumor Necrosis Factor-alpha/TNFSF2 (Canine TNF-α/TNFSF2)_C230273
Tumor necrosis factor alpha (TNF-alpha ), also known as cachectin and TNFSF2, is the prototypic ligand of the TNF superfamily. It is a pleiotropic molecule that plays a central role in inflammation, immune system development, apoptosis, and lipid metabolism. Canine TNF-alpha consisits of a 35 amino acid (aa) cytoplasmic domain, a 21 aa transmembrane segment, and a 177 aa extracellular domain (ECD). Within the ECD, canine TNF-alpha shares 84% ‑ 94% aa sequence identity with equine, feline, human, porcine, and rhesus and 69%-77% with bovine, cotton rat, mouse, and rat with TNF-alpha. TNF-alpha is produced by a wide variety of immune, epithelial, endothelial, and tumor cells. TNF-alpha is assembled intracellularly to form a noncovalently linked homotrimer which is expressed on the cell surface. Cell surface TNF-alpha can induce the lysis of neighboring tumor cells and virus infected cells, and it can generate its own downstream cell signaling following ligation by soluble TNFR I. Shedding of membrane bound TNF-alpha by TACE/ADAM17 releases the bioactive cytokine, a 55 kDa soluble trimer of the TNF-alpha extracellular domain. TNF-alpha binds the ubiquitous 55-60 kDa TNF RI and the hematopoietic cell-restricted 80 kDa TNF RII, both of which are also expressed as homotrimers. Both type I and type II receptors bind TNF-alpha with comparable affinity, although only TNF RI contains a cytoplasmic death domain which triggers the activation of apoptosis. Soluble forms of both types of receptors are released and can neutralize the biological activity of TNF-alpha. Recombinant Equine Interleukin-1 Receptor Antagonist Protein ( Equine IL-1RA)_C230277
Secreted equine IL-1 receptor antagonist (IL-1ra) is a presumably 22-25 kDa glycoprotein produced by variety of cell types that antagonizes IL-1 activity. It is a member of the IL-1 family of proteins that includes IL-1 alpha and IL-1 beta. Although there is little amino acid (aa) identity (< 30%) among the three IL-1 family members, all molecules bind to the same receptors, all show a beta -trefoil structure, and all are believed to have evolved from a common ancestral gene. Equine IL-1ra is synthesized as a 177 aa precursor that contains a 25 aa signal sequence plus a 152 aa mature region. There is one intrachain disulfide bond and one potential N-linked glycosylation site. Mature equine sIL-1ra is 78%, 78%, 80%, 82%, and 76% aa identical to mature mouse, human, porcine, canine and bovine IL-1ra, respectively. In human, three non-secreted IL-1ra isoforms have also been identified. It is unknown if such an analogous situation exists in equine. Cells known to secrete IL-1ra include fibroblasts, vascular smooth muscle cells, intestinal columnar epithelium, chondrocytes, macrophages, mast cells, neutrophils and hepatocytes. There are two type I transmembrane glycoprotein receptors for IL-1ra. The first is the bioactive 80 kDa type I IL-1 receptor (IL-1 RI), and the second is the inert (decoy) 65 kDa type II IL-1 receptor. IL-1ra binding to IL-1 RI competitively blocks IL-1 ( alpha or beta ) binding to the same receptor. This results in receptor ligation without activation. The type II IL-1 receptor is inert, and any binding of IL-1ra not only fails to block co-existing IL-1 activity, but may actually potentiate it by removing an IL-1 antagonist. Functionally, all activities attributed to IL-1ra are explained by its role as a competitive inhibitor of IL-1 binding to IL-1 RI. Recombinant Equine Interleukin-2 Cys141Ser ( Equine IL-2 Cys141Ser)_C230278
Interleukin 2 was initially identified as a T cell growth factor that is produced by T cells following activation by mitogens or antigens. IL-2 has since been found to also stimulate the growth and differentiation of B cells, natural killer (NK) cells, lymphocyte activated killer (LAK) cells, monocytes/macrophages and oligodendrocytes. The biological activity of IL-2 is mediated by the binding of IL-2 to cell surface receptor complexes. The functional high-affinity receptor that mediate IL‑2 signals is composed of three polypeptide chains, the IL-2 receptor alpha, beta and gamma subunits. IL-2 also signals via the intermediate affinity receptor complex of the beta and gamma subunits. In T cells, the beta and gamma subunits are shared with the IL-15 receptor complex. The gamma subunit of the IL-2 receptor complex has also been shown to be a subunit of the receptor complexes of IL-4, IL-7, IL-9 and IL-21. At the amino acid sequence level, equine IL-2 shares 72%, 70%, 56% and 54% sequence similarities with human, porcine, rat and mouse IL-2, respectively. It has been reported that equine IL-2 augmented proliferation in equine peripheral blood mononuclear cells, but has no effect on mouse CTLL-2 cells. Recombinant Human Cyclophilin D (Human CyP-D)_C230318
The protein encoded by this gene is a member of the peptidyl-prolyl cis-trans isomerase (PPIase) family. PPIases catalyze the cis-trans isomerization of proline imidic peptide bonds in oligopeptides and accelerate the folding of proteins. This protein is part of the mitochondrial permeability transition pore in the inner mitochondrial membrane. It is proposed that its association with the mPTP is masking a binding site for inhibiting inorganic phosphate (Pi) and promotes the open probability of the mPTP leading to apoptosis or necrosis; the requirement of the PPIase activity for this function is debated. In cooperation with mitochondrial p53/TP53 is involved in activating oxidative stress-induced necrosis. Involved in modulation of mitochondrial membrane F(1)F(0) ATP synthase activity and regulation of mitochondrial matrix adenine nucleotide levels. Has anti-apoptotic activity independently of mPTP and in cooperation with BCL2 inhibits cytochrome c-dependent apoptosis. Dkk proteins are secreted proteins that contain two conserved cysteine-rich domains separated by a linker region. Each domain contains ten cysteine residues . Mature human Dkk-1 is a 40 kDa glycosylated protein that shares 86%, 87%, 90% and 91% aa sequence identity with mouse, rat, rabbit and bovine Dkk-1, respectively. It also shares 42% and 36% aa identity with human Dkk-2 and Dkk-4, respectively. Dkk-1 and Dkk-4 are well documented antagonists of the canonical Wnt signaling pathway. This pathway is activated by Wnt engagement of a receptor complex composed of the Frizzled proteins and one of two low-density lipoprotein receptor-related proteins, LRP5 or LRP6 . Dkk-1 antagonizes Wnt by forming ternary complexes of LRP5/6 with Kremen1 or Kremen2. Dkk- 1/LRP6/Krm2 complex internalization has been shown to down-regulate Wnt signaling. Dkk-1 is expressed throughout development and antagonizes Wnt-7a during limb development. Other sites of expression include developing neurons, hair follicles and the retina of the eye . Product Properties Synonyms Dickkopf-1;DKK-1;SK Protein; DKK1; dkk1; dkk-1 Accession O94907 Source HEK293 Cells-derived human Dkk-1 protein, Met2-His 266 with His tag at the C-terminus. Recombinant Human Desert Hedgehog Cys23IleIle (Human DHH C23II)_C230320
Desert Hedgehog (Dhh) belongs to the highly conserved Hedgehog family of proteins which are involved in multiple developmental processes. Desert Hedgehog is a secreted, 45 kDa, 373 amino acid (aa) protein that undergoes autocatalytic cleavage between Gly198 and Cys199 catalyzed by the C-terminal domain, which releases the N-terminal domain with a concominant attachment of cholesterol at its new C-terminus. In addition to the C-terminally attached cholesterol, a fatty acid acyl chain is esterified to the N-terminal cysteine (aa 23) via an amide linkage. The 19 kDa N-terminal signaling domain is membrane associated due to its double lipid modifications. Its binding to Patched receptors results in the loss of Patched repression of Smoothened signaling. Dhh binds both Patched and Patched 2 as well as Hedgehog interacting protein (Hip). Within the N-terminal domain, human Dhh shares 97% aa sequence identity with mouse and rat Dhh. It shares approximately 75% aa seqeuence identity with human Indian (Ihh) and Sonic Hedgehog (Shh). Dhh is produced by Sertoli cells and is required for testis development and spermatogenesis. It induces steroidogenic factor 1, which is instrumental in promoting Leydig cell differentiation. It also promotes the deposition of basal lamina surrounding seminiferous tubules. Mutations of Dhh are linked to complete pure gonadal dysgenesis. Dhh is expressed in the female by ovarian granulosa cells and the corporus luteum. Its upregulation in ovarian cancer correlates positively with proliferative index, and negatively with prognosis. Dhh is also expressed by Schwann cells and is upregulated following nerve injury. It induces the expression of Patched and Hip in neural tissue fibroblasts and promotes the formation of the connective tissue sheath surrounding peripheral nerves. Recombinant Human Eotaxin-3/CCL26 (Human Eotaxin-3/CCL26)_C230327
Eotaxin-3, also named CCL26 or SCYA26, is a novel human CC chemokine. The eotaxin subfamily of CC chemokines consists of eotaxin-1/CCL11, eotaxin-2/CCL24 and eotaxin-3/CCL26. Eotaxin-3/CCL26 has been shown to be expressed by several different cell types, including basophils, dendritic cells, smooth muscle cells, epithelial cells and fibroblasts. Interleukin (IL)-4 is the main inducer for eotaxin-3 expression, whereas eotaxin-1 is up-regulated by IL-4 and the proinflammatory cytokine tumour necrosis factor (TNF)-α. Eotaxin-3 is expressed in vascular endothelial cells and human dermal fibroblasts after IL-4 and IL-13 stimulation, and this is dependent upon the IL-4-/IL-13-specific transcription factor, signal transducers and activator of transcription (STAT)-6. Eotaxin-3 is expressed on the surface of IL-4-stimulated endothelial cells and promotes eosinophil transmigration. Recombinant Human Epididymis Protein 4 (Human E4)_C230330
Epididymis Protein 4, also known as Epididymal secretory protein E4, Major epididymis-specific protein E4, Putative protease inhibitor WAP5, WFDC2 and HE4, is a secreted protein that contains two WAP domains. WFDC2 / HE4 is a member of a family of stable 4-disulfide core proteins that are secreted at high levels. It is expressed in a number of normal tissues, including male reproductive system, regions of the respiratory tract and nasopharynx. It is highly expressed in a number of tumors cells lines, such ovarian, colon, breast, lung and renal cells lines. Initially described as being exclusively transcribed in the epididymis. WFDC2 may be a component of the innate immune defences of the lung, nasal and oral cavities and suggest that WFDC2 functions in concert with related WAP domain containing proteins in epithelial host defence. WFDC2 re-expression in lung carcinomas may prove to be associated with tumour type and should be studied in further detail. Mammary gland expression of tammar WFDC2 during the course of lactation showed WFDC2 was elevated during pregnancy, reduced in early lactation and absent in mid-late lactation. Recombinant Human B-Cell Maturation Antigen/TNFRSF17 (Human BCMA/TNFRSF17)_C230299
Tumor necrosis factor receptor superfamily, member 17 (TNFRSF17), also known as B cell maturation antigen (BCMA) or CD269 antigen, is a member of the TNF receptor superfamily. It is a type III membrane protein containing one extracellular cysteine rich domain. This receptor is preferentially expressed in mature B lymphocytes, and may be important for B cell development and autoimmune response. TNFRSF17 can binds to TNFSF13B/BLyS/BAFF and TNFSF13/APRIL, and to lead to NF-kappaB and MAPK8/JNK activation. TNFRSF17 also binds to various TRAF family members, and thus may transduce signals for cell survival and proliferation. TNFRSF17 is a receptor for TALL-1 and BCMA activates NF-kappaB through a TRAF5-, TRAF6-, NIK-, and IKK-dependent pathway. The identification of TNFRSF17 as a NF-kappaB-activating receptor for TALL-1 suggests molecular targets for drug development against certain immunodeficient or autoimmune diseases. TNFRSF17/BCMA is a target of donor B-cell immunity in patients with myeloma who respond to DLI. Antibody responses to cell-surface BCMA may contribute directly to tumor rejection in vivo. Mature human CCL27 is an 88 amino acid (aa) protein that shares 57% aa sequence identity with mouse and rat CCL27. It shares 11% - 35% aa sequence identity with other human CC chemokines. An alternately spliced form of mouse CCL27, known as PESKY, is localized to the nucleus and promotes cellular migration. CCL27 is constitutively expressed by keratinocytes and is upregulated by inflammatory stimuli and in wounded skin. CCL27 binds the chemokine receptor CCR10, glycosaminoglycans in the extracellular matrix, sulfated tyrosine residues on PSGL-1, and determinants on the surface of fibroblasts and endothelial cells . CCL27 cooperates with CCL17/TARC in inducing the migration of cutaneous lymphocyte antigen (CLA) positive memory T cells to the skin during inflammation. Endothelial cell-bound CCL27 can mediate the adhesion of those cells to CLA+ T cells. CCL27 also induces the migration of keratinocyte precursors from bone marrow to the skin, thereby promoting wound healing. In humans, serum CCL27 levels are elevated and correlate with disease severity in atopic dermatitis, psoriasis vulgaris, and mycosis fungoides. Recombinant Human Acidic Fibroblast Growth Factor, 2-155aa (Human aFGF,2-155aa)_C230287
FGF acidic, also known as FGF1, ECGF, and HBGF-1, is a 17 kDa nonglycosylated member of the FGF family of mitogenic peptides. FGF acidic, which is produced by multiple cell types, stimulates the proliferation of all cells of mesodermal origin and many cells of neuroectodermal, ectodermal, and endodermal origin. It plays a number of roles in development, regeneration, and angiogenesis. Human FGF acidic shares 54% amino acid sequence identity with FGF basic and 17%- 33% with other human FGFs. It shares 92%, 96%, 96%, and 96% aa sequence identity with bovine, mouse, porcine, and rat FGF acidic, respectively, and exhibits considerable species crossreactivity. Alternate splicing generates a truncated isoform of human FGF acidic that consists of the N-terminal 40% of the molecule and functions as a receptor antagonist. During its nonclassical secretion, FGF acidic associates with S100A13, copper ions, and the C2A domain of synaptotagmin 1. It is released extracellularly as a disulfide-linked homodimer and is stored in complex with extracellular heparan sulfate . The ability of heparan sulfate to bind FGF acidic is determined by its pattern of sulfation, and alterations in this pattern during embryogenesis thereby regulate FGF acidic bioactivity . The association of FGF acidic with heparan sulfate is a prerequisite for its subsequent interaction with FGF receptors. Recombinant Human Angiostatin (Human Angiostatin)_C230290
Angiostatin is an anti- angiogenic 38- 45 kDa proteolytic fragment of Plasminogen, a 92- 100 kDa glycosylated blood zymogen that serves as the precursor for Plasmin. Plasminogen is produced primarily in the liver, but also in other tissues. Angiostatin circulates in the plasma, binds endothelial cells and myeloid cells, is present in platelet granules, and is excreted in the urine. Human Plasminogen contains an N-terminal activation peptide between amino acids (aa) 1- 98, five characteristically folded kringle domains (aa 103- 561), and a peptidase S1 domain (aa 581- 808). Cleavage of the activation peptide produces mature Plasminogen, while further cleavage between Arg580 and Val581 by tPA (tissue plasminogen activator) produces the disulfide-linked two-subunit enzyme plasmin that dissolves fibrin clots. Angiostatin was first identified as consisting of kringles 1- 4, a form called K1- 4. Human Angiostatin (K1- 4, aa 99- 459) shares 79- 80% aa sequence identity with mouse, rat, canine, feline, porcine and bovine K1- 4. Other anti- angiogenic forms include kringles 1- 3 (K1- 3), or 1- 4 plus most of kringle 5 (K4.5). K4.5, which is reported to be the most active form, occurs in vivo by autoproteolysis of mature Plasminogen in the presence of either a sulfhydryl donor or cell surface actin, while matrix metalloproteins such as MMP3, 7, 9 and 19 can create multiple forms. Recombinant Human Betacellulin (Human Betacellulin)_C230301
Betacellulin (BTC) is a new member of the EGF family of cytokines that is comprised of at least ten proteins including EGF, TGF-alpha, amphiregulin, HB-EGF, and the various heregulins. All of these cytokines are synthesized as transmembrane precursors and are characterized by the presence of one or more EGF structural units in their extracellular domain. The soluble forms of these cytokines are released by proteolytic cleavage. BTC, a heparin-binding protein, was originally isolated from the conditioned media of mouse pancreatic beta tumor cells as a 32 kDa glycoprotein composed of 80 amino acid residues. The cDNA encoding human BTC was cloned from a human breast adenocarcinoma cell line (MCF-7) cDNA library. Human and mouse cDNAs encode BTC precursor proteins of 178 and 177 amino acid residues, respectively. At the amino acid sequence level, human BTC precursor protein exhibits 79% identity with that of the mouse BTC precursor. In a mouse cell line transfected with human BTC cDNA, three forms of soluble human BTC have been detected: the glycosylated, intact BTC composed of 80 amino acid residues, a truncated molecule lacking 12 amino acid residues from the amino terminus, and a second truncated molecule lacking 30 amino acid residues from the amino terminus. The biological activities of these BTC forms were shown to be identical. BTC can bind to the EGF receptor and is a potent mitogen for Balb/c 3T3 fibroblasts, retinal pigment epithelial cells and vascular smooth muscle cells. Recombinant Flagellin, His ( Flagellin, His)_C230280
The role of flagella and motility in the attachment of the foodborne pathogen Listeria monocytogenes to various surfaces is mixed with some systems requiring flagella for an interaction and others needing only motility for cells to get to the surface. In nature this bacterium is a saprophyte and contaminated produce is an avenue for infection. Previous studies have documented the ability of this organism to attach to and colonize plant tissue. Motility mutants were generated in three wild type strains of L. monocytogenes by deleting either FlaA, the gene encoding flagellin, or motAB, genes encoding part of the flagellar motor, and tested for both the ability to colonize sprouts and for the fitness of that colonization. The motAB mutants were not affected in the colonization of alfalfa, radish, and broccoli sprouts; however, some of the FlaA mutants showed reduced colonization ability. The best colonizing wild type strain was reduced in colonization on all three sprout types as a result of a FlaA deletion. A mutant in another background was only affected on alfalfa. The third, a poor alfalfa colonizer was not affected in colonization ability by any of the deletions. Fitness of colonization was measured in experiments of competition between mixtures of mutant and parent strains on sprouts. Here the FlaA and motAB mutants of the three strain backgrounds were impaired in fitness of colonization of alfalfa and radish sprouts, and one strain background showed reduced fitness of both mutant types on broccoli sprouts. Together these data indicate a role for flagella for some strains to physically colonize some plants, while the fitness of that colonization is positively affected by motility in almost all cases. Recombinant Human Epithelial Neutrophil Activating Peptide-78, 5-78 aa/CXCL5 (Human ENA-78,5-78 aa/CXCL5)_C230333
CXCL5, also known as epithelial cell-derived neutrophil-activating peptide (ENA-78), is an 8 kDa proinflammatory member of the CXC chemokine family. Its Glu-Leu-Arg (ELR) motif confers angiogenic properties and distinguishes it from ELR-CXC chemokines which are angiostatic. CXCL5 is produced following stimulation of cells with the inflammatory cytokines interleukin-1 or tumor necrosis factor-alpha. It also can be detected in eosinophils, and can be inhibited with the type II interferon. In vivo, CXCL5 is elevated at sites of inflammation and pulmonary fibrosis where it promotes neutrophil infiltration and activation as well as angiogenesis. Its up-regulation contributes to increased vascularization, tumor growth, and metastasis in many cancers. CXCL5 plays a role in reducing sensitivity to sunburn pain in some subjects, and is a potential target which can be utilized to understand more about pain in other inflammatory conditions like arthritis and cystitis. It elicits these effects by interacting with the cell surface chemokine receptor CXCR2. Recombinant Human Erythropoietin ,Fc (Human EPO,Fc)_C230334
Erythropoietin (EPO) is a 34 kDa glycoprotein hormone in the type I cytokine family and is related to thrombopoietin. Its three N-glycosylation sites, four alpha helices, and N- to C-terminal disulfide bond are conserved across species. Glycosylation of the EPO protein is required for biological activities in vivo . The mature human EPO protein shares 75% - 84% amino acid sequence identity with bovine, canine, equine, feline, mouse, ovine, porcine, and rat EPO. EPO is primarily produced in the kidney by a population of fibroblast-like cortical interstitial cells adjacent to the proximal tubules. It is also produced in much lower, but functionally significant amounts by fetal hepatocytes and in adult liver and brain. EPO promotes erythrocyte formation by preventing the apoptosis of early erythroid precursors which express the erythropoietin receptor (EPO R). EPO R has also been described in brain, retina, heart, skeletal muscle, kidney, endothelial cells, and a variety of tumor cells . Ligand induced dimerization of EPO R triggers JAK2-mediated signaling pathways followed by receptor/ligand endocytosis and degradation. Rapid regulation of circulating EPO allows tight control of erythrocyte production and hemoglobin concentrations. Recombinant Human Exodus-2/CCL21 (Human Exodus-2/CCL21)_C230335
CCL21, also known as 6Ckine, TCA-4, SLC, Exodus-2, and A21, is a 12 kDa homeostatic chemokine. Some of the chemokines are considered pro-inflammatory which can be induced to recruit cells of the immune system to a site of infection during an immune response, while others are considered homeostatic and are implied in controlling the migration of cells during normal processes of tissue maintenance and development. CCL21 takes its name 6Ckine for its consititutively six conserved cysteine residues but not four cysteines typical to chemokines. CCL21 has function in ininducing vigorous calcium migrations and chemotactic responses. During chronic inflammation or tissue damage, CCL21 is up-regulated on local vascular endothelial cells, macrophages, T cells, and neurons. In these settings, it promotes fibrosis, inflammatory cytokine production, and neuropathic pain. The soluble chemokine is elevated in rheumatoid arthritis synovial fluid and in the serum of coronary artery disease patients. CCL21 has been shown to exert either angiogenic or angiostatic effects. These effects, in combination with the ability of CCL21 to attract immune suppressor cells (Treg and MDSC) to a tumor site can have positive or negative effects on tumor progression.
